I would imagine a few months. Given the circumstance, it might just go to the house of Representatives to decide who should be president while an election is put together. FTR: it's never happened in US History.

It's hard to imagine hearing on the news that all political leaders were killed in one explosion. But we wouldn't know exactly what would happen right?

That's about 20 people you'd have to kill. With their own agendas (and probably) high security, it'd be hard to pull of. And if one cabinet member goes down, I'm sure the rest would be put on high alert.
